Il s'intéresse au théâtre pendant ses études universitaires au San Francisco college et fait son apprentissage au "Black actors' workshop" de l'"American conservatory theater". En 1979, Glover fait ses débuts au cinéma aux côtés de Clint Eastwood dans "L'évadé d'Alcatraz" de Don Siegel.
